Bioavailability of Citrulline in Watermelon Flesh, Rind, and Skin Using a Human Intestinal Epithelial Caco-2 Cell Model

چکیده

Watermelon produces many byproducts (watermelon rind and skin) even though those components contain various bioactive compounds, including citrulline. This study evaluated the citrulline concentration, total phenolic content, antioxidant activity (DPPH FRAP assays) of different parts watermelon investigated bioavailability from using an in vitro human intestinal epithelial Caco-2 cell monolayer model. Solid-phase extracted flesh, rind, skin samples were treated on a for 1, 2, 4 h. The collected basolateral solution at each time point was analyzed percentage transport. flesh had highest but content compared to other parts. showed greater % transport than L-citrulline standard. It may be due food matrices suggests that utilization by-products such as would help develop value-added products with better However, since this conducted model, more extensive research vivo studies will needed.

Determination of citrulline in watermelon rind.

Watermelon (Citrullus vulgaris Schrad.) is a natural and rich source of the non-essential amino acid citrulline. Citrulline is used in the nitric oxide system in humans and has potential antioxidant and vasodilatation roles. A method using gas chromatography-mass spectrometry (GC-MS) was developed to separate citrulline from glutamic acid, which co-elute when analyzed by high performance liquid...

Qualitative inheritance of rind pattern and flesh color in watermelon.

Watermelon [Citrullus lanatus (Thunb.) Matsum. & Nakai var. lanatus] is a diverse species, with fruits of different sizes, shapes, rind patterns, and flesh colors. This study measured the inheritance of novel rind phenotypes and verified the genetics of white, red, salmon yellow, and canary yellow flesh colors. For each of the 11 crosses, six generations (P(a)S1, P(b)S1, F1, F2, BC1P(a), and BC...

Evaluation of Iron Bioavailability in Caco-2 cell Culture Model: Modification of the Original Method

Background and Objectives: In the original in vitro method for evaluation of iron bioavailability using caco-2 cell model, ferritin formation by cells is considered as a functional indicator of both absorption and utilization of iron. However, as iron is measured in the tested material and then ferritin concentration is measured in caco-2 cell lysate, it is almost impossible to have a concrete ...

Intestinal Epithelial Model - the Caco-2 Cell Monolayer

The absorption of orally administered drugs requires their movement across the intestinal epithelial barrier. Poor intestinal permeability leads to limited absorption. Generally, if a compound achieves a 90% or greater oral absorption, it is considered highly permeable (for example, propranolol or metoprolol). Drugs that display 50% or less oral absorption are considered poorly permeable (for e...

Transport of thiamine in human intestine: mechanism and regulation in intestinal epithelial cell model Caco-2.

The present study examined the intestinal uptake of thiamine (vitamin B1) using the human-derived intestinal epithelial cells Caco-2 as an in vitro model system. Thiamine uptake was found to be 1) temperature and energy dependent and occurred with minimal metabolic alteration; 2) pH sensitive; 3) Na+ independent; 4) saturable as a function of concentration with an apparent Michaelis-Menten cons...
